Kit 4: Preparation of Human Chromosome Spreads

Using this kit, each student will be provided the unique opportunity of preparing a chromosome spread using human cells. The excitement generated by the students as they "splat" the cells onto microscope slides and proceed to "hunt" for the ideal chromosome spread with the microscope has generated an equal level of excitement among the hundreds of teachers who use this kit. Because the cells used for this exercise are transformed, the number of chromosomes will vary among the cells. Each student can count the chromosomes and see firsthand the range of aneuploidy.
The manual provides information regarding chromosome structure and the important role of karyotyping in geneticanalysis and genetic disease diagnosis.

Kit includes
  • Enough materials for 30 students working individually.
  • 15 tubes each containing metaphase-blocked cell suspension, fixed in acetic acid-methanol fixative.

Teacher's Preparation

  1. Binocular or monocular microscopes - magnification of 400X to 1000X (oil immersion) will enable the students to identify and count chromosomes ans see sister chromatids.
  2. Pipettes, microscope slides, coverslips.
  3. Staining jars or beakers ( = 250ml) containing room temperature water.

Time Requirements
40-60 minutes.
